gEL-DNA: een klonerings- en polymerasekettingreactievrije methode voor op CRISPR gebaseerde gemultiplexte genoombewerking
Zelfs voor de genetisch toegankelijke gist Saccharomyces cerevisiae heeft de CRISPR-Cas DNA-bewerkingstechnologie de stamconstructie sterk versneld en vergemakkelijkt. Verschillende methoden zijn gevalideerd voor snelle en zeer efficiënte enkelvoudige bewerkingsgebeurtenissen, en verschillende benaderingen voor multiplex genoombewerking zijn in de literatuur beschreven door middel van Sp Cas9- of Fn Cas12a-endonucleasen en de bijbehorende gids-RNA’s (gRNA’s). De gRNA’s die worden gebruikt om het Cas-endonuclease naar de bewerkingssite te leiden, worden meestal uitgedrukt uit plasmiden met behulp van inheemse Pol II- of Pol III-RNA-polymerasen.
Deze gRNA-expressieplasmiden vereisen moeizame, tijdrovende kloonstappen , wat de implementatie ervan voor academische en toegepaste doeleinden belemmert. In deze studie onderzoeken we het potentieel van het tot expressie brengen van gRNA uit lineaire DNA-fragmenten met behulp van de T7 RNA-polymerase (T7RNAP) voor enkelvoudige en multiplex genoombewerking in Saccharomyces cerevisiae . Met behulp van Fn Cas12a toont dit werk aan dat het transformeren van korte, lineaire DNA-fragmenten die coderen voor gRNA’s in giststammen die T7RNAP tot expressie brengen, zeer efficiënte enkelvoudige en duplex DNA-bewerking bevordert. Deze DNA-fragmenten kunnen op maat worden besteld, waardoor deze benadering zeer geschikt is voor de constructie van stammen met een hoge doorvoer. Dit werk breidt de CRISPR . uittoolbox voor grootschalige stamconstructieprogramma’s in S. cerevisiae en belooft related te zijn voor andere minder genetisch toegankelijke gistsoorten.
Klonen van menselijke T-cellen door verdunning te beperken
Menselijke T-cellen vertegenwoordigen een heterogene populatie , inclusief cellen met verschillende fenotypische en functionele eigenschappen. Ondanks dat er de afgelopen jaren verschillende technologieën zijn ontwikkeld om fenotypische eigenschappen van T-cellen op eencellig niveau te onderzoeken, blijft de in vitro T-celklooncultuur de enige manier om functioneel onderzoek uit te voeren op T-cellen op eencellig niveau. Hier beschrijven we de methode om menselijke T-celklonen te verkrijgen door de verdunning in de aanwezigheid van feedercellen te beperken en ze in cultuur te houden voor verder onderzoek.
Biosynthese van meervoudig onverzadigde vetzuren met lange keten in mariene gammariden: moleculaire klonering en functionele karakterisering van drie vette acyl-elongasen
Langketenige (C 20-24 ) meervoudig onverzadigde vetzuren (LC-PUFA’s) zijn essentiële voedingsstoffen die meestal worden geproduceerd in mariene ecosystemen. Eerdere research suggereerden dat gammariden enig vermogen hebben om endogeen LC-PUFA’s te produceren. Deze studie had tot doel het repertoire en de functies van verlenging van zeer lange-keten vetzuur (Elovl) eiwitten in gammariden te onderzoeken. Onze resultaten laten zien dat gammariden ten minste drie verschillende elovl- genen hebben met vermeende rollen in de biosynthese van LC-PUFA. Fylogenetica stelde ons in staat om twee elongasen te classificeren als Elovl4 en Elovl6, omdat ze bonafide orthologen waren van gewervelde Elovl4 en Elovl6. Bovendien werd een derde elongase genoemd als “Elovl1/7-achtig” omdat het nauw gegroepeerd was met de Elovl1 en Elovl7 die worden aangetroffen in gewervelde dieren.
Moleculaire analyse van de afgeleide eiwitsequenties gaf aan dat de gammariden Elovl4 en Elovl1/7-achtige inderdaad meervoudig onverzadigde vetzuur (PUFA) elongasen waren, terwijl Elovl6 moleculaire kenmerken had die typisch gevonden worden in niet-PUFA elongasen. Dit werd gedeeltelijk bevestigd in de functionele assays uitgevoerd op het mariene vlokreeft Echinogammarus marinus Elovl, waaruit bleek dat zowel Elovl4 en Elovl1 / 7-PUFA soortgelijke langgerekte substraten variëren van C 18 tot C 22 . E. marinus Elovl6 kon slechts C langwerpige 18 PUFA substraten, wat suggereert dat dit enzym geen belangrijke rol speelt bij de LC-PUFA biosynthese van vlokreeftjes.

Klonen , expressie, karakterisering en antioxidantbescherming van glutaredoxine3 tegen psychrofiele bacterie Psychrobacter sp. ANT206
Glutaredoxines (Grxs) zijn eiwitten die de glutathion (GSH)-afhankelijke reductie van eiwitdisulfiden katalyseren. In deze studie werd een Grx-gerelateerd gen (264 bp) , dat codeert voor een Ps-Grx3, gekloneerd uit Psychrobacter sp. ANT206. Sequentieanalyse duidde op de aanwezigheid van het actieve plaatsmotief CPYC in dit eiwit. Homologiemodellering toonde aan dat Ps-Grx3 minder waterstofbruggen en zoutbruggen had, evenals een lagere Arg/(Arg + Lys)-verhouding dan zijn mesofiele homologen, wat wijst op een verbeterd katalytisch vermogen bij lage temperaturen. Plaatsgerichte mutagenese toonde aan dat de Cys13, Professional14 en Cys16websites waren essentieel voor de katalytische activiteit van Ps-Grx3, terwijl circulair dichroïsme (CD) spectroscopie bevestigde dat puntmutaties in deze aminozuurresiduen leidden tot het verlies of de vermindering van enzymactiviteit.
Verder toonde analyse van de biochemische eigenschappen van Ps-Grx3 aan dat de optimale temperatuur van dit enzym 25°C was. Belangrijk is dat Ps-Grx3 gevoeliger was voor tBHP en CHP dan voor H 2 O 2 , en ongeveer 40% activiteit behield, zelfs wanneer de H 2 O 2 -concentratie werd verhoogd tot 1 mm. Wat betreft substraatspecificiteit had Ps-Grx3 een hogere affiniteit voor HED, L-cystine en DHA dan voor S-sulfocysteïne en BSA. We onderzochten ook het DNA-beschermende vermogen van Ps-Grx3 met behulp van het pUC19-plasmide en ontdekten dat Ps-Grx3 supercoiled DNA kon beschermen tegen oxidatie-geïnduceerde schade bij 15 ° C gedurende 1,5 uur. Moleculair klonen en functionele research van magang goose toll-like receptor
Uitbraken van ziekten hebben een grote invloed op de economische levensvatbaarheid van de dierindustrie. Er is weinig bekend over de mechanismen van aan het immuunsysteem gerelateerde ziekten bij ganzen. Toll-like receptoren (TLR’s) spelen bij de meeste diersoorten een belangrijke rol in het ontstekingsremmende immuniteitsproces, maar zijn niet onderzocht bij de Magang-gans. Om de rol van TLR’s op te helderen, werden reverse transcriptie-polymerasekettingreactie (RT-PCR) en PCR-amplificatie van cDNA-uiteinden (Sensible RACE) gebruikt om het Magang-gans TLR5-gen (mgTLR5) te klonen. Het cDNA van volledige lengte van mgTLR5 was 2967bp lang, inclusief een 5′-terminaal onvertaald gebied (UTR) van 215 bp, een 3′-terminaal UTR van 384 bp en een open leesraam van 2583 bp dat codeert voor een eiwit van 860 aminozuren. Structureel gezien heeft mgTLR5 een toll/interleukine-receptor (TIR) domein, een transmembraan domein en zeven leucine-rich repeats (LRR’s) domeinen.
Homologie-uitlijning van TLR5 en zijn TIR- domeinen met andere soorten onthulde dat mgTLR5 98% en 81,3% van de sequentieovereenkomst deelde met respectievelijk witte gans TLR5 en kippen TLR5. Kwantitatieve RT-PCR toonde aan dat het mgTLR5-gen van de gans breed tot expressie wordt gebracht in alle geteste weefsels, met de hoogste expressie in de nier en de milt. De toename in NF-KB-promoteractiviteit gestimuleerd door flagelline was afhankelijk van mgTLR5-expressie in 293 T-cellen. Salmonella pullorum en flagellinede expressie van TLR5-, IL-8- en IL-1-mRNA in perifere bloedmononucleotidecellen van in vitro gekweekte Magang-ganzen aanzienlijk opgereguleerd. Stimulatie door S. pullorum gedurende 24 uur verhoogde de expressie van mgTLR5 in de blindedarm en de nieren. We concluderen dat Magang-gans TLR5 een functionele TLR5-homoloog is van het eiwit in andere soorten en een belangrijke rol speelt bij bacteriële herkenning.
